A review of frequentist methods for combining results of series of trials

چکیده
Background: A randomized controlled trial is considered the gold standard in clinical research, also in rare diseases. However, in small populations, single large-scale well-powered trials are often not possible. For valid decision-making, both on efficacy and on safety, evidence generated from series of trials could be exploited. Methods: We conducted a review over a five-year period between January 2009 and December 2013 to identify relevant methodology on combining results of series of trials. Results: Sixty-two papers were identified and summarized in this review. Its focus is on frequentist methodology. Most papers deal with meta-analyses on aggregated data. Only few papers discuss multivariate outcomes. We categorized the relevant methods according to the type of (meta-) analysis. Conclusions: Only a few papers dealt directly with series of trials in small populations. The results of the review lead to some directions for further investigation on evidence-based decision-making from a (small) number of trials in small populations.

A mixed Bayesian/Frequentist approach in sample size determination problem for clinical trials
In this paper we introduce a stochastic optimization method based ona mixed Bayesian/frequentist approach to a sample size determinationproblem in a clinical trial. The data are assumed to come from a nor-mal distribution for which both the mean and the variance are unknown. In contrast to the usual Bayesian decision theoretic methodology, whichassumes a single decision maker, our method recogni...
متن کاملRefractory iron deficiency anemia and Helicobacter Pylori Infection in pediatrics: A review
Background Since the discovery of Helicobacter pylori, several clinical reports have demonstrated that H. Pylori infection has emerged as a new cause of refractory iron stores in children. We carried out a systematic literature review to primarily evaluate the existing evidence on the association between childhood H. Pylori infection and iron deficiency anemia (IDA) and secondly, to investigat...
متن کاملActivity of Nitazoxanide Against Viral Gastroenteritis: A Systematic Review
Introduction: Nitazoxanide is an oral anti-parasitic agent that has been found to have broad antiviral activity. Its role in the treatment of viral gastroenteritis is not well-studied. Given the worldwide prevalence of viral gastroenteritis, particularly in developing nations, a systematic review of this topic would be valuable. Methods: A formal literatu...

The paper presents unified analytical solution for combining high-frequency and low-frequency economic time-series by additive and proportional Denton methods with parametrical dependence on the initial values of variable and indicator in evident form. This solution spans Denton’s original and Cholette’s advanced benchmarking initial conditions as the subcases. Computational complexity of the o...
متن کاملComparison of Bayesian and Frequentist Methods in Estimating the Net Reclassification and Integrated Discrimination Improvement Indices for Evaluation of Prediction Models: Tehran Lipid and Glucose Study
Introduction: The Frequency-based method is commonly used to estimate the Net Reclassification Improvement (NRI)- and Integrated Discrimination Improvement (IDI) indices. These indices measure the magnitude of the performance of statistical models when a new biomarker is added. This method has poor performance in some cases, especially in small samples. In this study, the performance of two Bay...
